Chronos is 42 minutes long and has no actors or dialogue. The soundtrack consists of a single continuous piece by composer Michael Stearns. Filmed in dozens of locations on five continents, the film relates to the concept of time passing on different scales—the bulk of the film covers the history of civilization, from pre-history to Egypt to Rome to Late Antiquity to the rise of Western Europe in the Middle Ages to the Renaissance to the modern era. It centers on European themes but not exclusively. Other time scales include the passing of seasons, and the passing of night and day, and the passing shadows of the sun in an afternoon to the passing of people on the street. These themes are intermingled with symbolic meaning.

This musical drama sweeps through a turbulent 40 years in popular singer Marika's (Sotiria Leonardou) life – and in the history of Greece – starting with the singer's birth in Smyrna, Turkey in 1917. Marika was deported to Greece along with all the other Greeks in Smyrna when she was seven years old, and a few years later, her parents started a career as a musician and a singer in a nightclub/bar. In the short space of one decade, Marika witnesses her father murder her mother, runs away from home, has a baby, and comes back to the nightclub to sing in an act with a childhood friend and a bouzouki player, Babis. Success finally comes at the expense of the suicide of another female singer at the club (named Rosa), but then Marika's childhood friend is exiled for political reasons, and she and Babis leave for other venues. Although Ninou carries a torch for Babis, their relationship never seems to work out, and after many years and World War II go by, she sends her daughter away to a convent school and goes on a tour in America. She then returns to Greece to find herself supplanted by a younger singer named Matina who has caught the attention Babis. Marika eventually dies in an accident during Mardi Gras and is buried by the people she sang with as they sing in her memory at the cemetery.

The concept of the movie is explained in a voiceover intro by G. W. Bailey, who wonders what it would be like if one of the 1930s/1940s Rex O'Herlihan movies were to be made today. At that point, in a scene reminiscent of The Wizard of Oz, the cinematography shifts from black & white to color and the soundtrack changes from mono to Dolby Digital surround sound.
