Theresa Jones (Zoe Saldana) takes her boyfriend, Simon Green (Ashton Kutcher) home to meet her parents on the occasion of her parents' 25th wedding anniversary, planning to reveal that they are engaged. However, what Theresa has also left out is that Simon is white. Her father, Percy (Bernie Mac), dislikes Simon almost immediately because of his race (Joneses are black). Wishing to impress Percy, Simon lies to him about being a NASCAR Pit driver for Jeff Gordon, not realizing that Percy is one of his biggest fans. After catching Theresa and Simon in a compromising position, Percy tries to force Simon into a hotel, but all the hotels in town are booked. Instead, he allows Simon to sleep in his basement on the couch, where Percy also sleeps.

Jack Slavin (Daniel Day-Lewis), a Scottish farmer with a heart illness, lives on an island which had been a hippie commune decades before. He is struggling to keep landowners from building developments on the wetland. His teenaged daughter Rose (Camilla Belle) is a beautiful but isolated girl with a passion for gardening. Since Rose's mother had left the family, Jack homeschooled his daughter and did not expose her to life beyond their small island home. Jack believes that they both "need a woman around." The plot focuses on six women known as thralls, half-vampires who lack the ability to fly or turn their victims. The thralls are joined by Ashley (Baruc), the sister of one of the thralls, and together they attempt to escape from the control of Mr. Jones (Lamas), a centuries-old vampire with a henchman named Rennie. Leslie opens a dance club in Iowa, while waiting the arrival of her sister Ashley, who grew up in an abusive household. Her father dies of a heart attack, causing Ashley to live with her sister.

Astronaut and Osteopath, Dr. Ivan Hood (Bruce Campbell) and his companion Kelly (Renee O'Connor) return to Earth after a 40 year space mission with two other astronauts. They are soon captured by other humans and one of their team is killed after sustaining a sprain; Dr. Hood pointed out that it would have healed in a few days. They are taken to a work camp, to their surprise, is run by locust-like aliens. These aliens had arrived 20 years ago to feast on the trees and used E.M.P. bombs to cripple earth before their invasion. The other member of their team is eaten alive by one of the aliens after refusing to work. The camp is supervised by human bounty hunters; they are treated better. After seeing how illiterate and savage humans have become due to the aliens, Dr. Hood and Kelly plan to make an escape the next day after hearing rumors that the President has survived and has an army waiting to strike back.

Four coeds are about to graduate from a secret paramilitary academy known as D.E.B.S. (Discipline, Energy, Beauty, and Strength). The team consists of Max Brewer (Meagan Good), who tends to draw her gun at the slightest provocation, Janet (Jill Ritchie), who wishes for nothing but to earn her stripes as a true D.E.B., the chain-smoking and promiscuous Dominique (Devon Aoki), and Amy Bradshaw (Sara Foster), who is doubtful about being a spy, despite being an honor student at the academy. Amy has broken up with her boyfriend Bobby Matthews (Geoff Stults), a Homeland Security agent. Her personal life takes a back seat to the re-emergence of Lucy Diamond (Jordana Brewster), a criminal mastermind who, by coincidence, is the subject of Amy's thesis. The D.E.B.S. are sent by the heads of D.E.B.S., Mrs. Petrie (Holland Taylor) and Mr. Phipps (Michael Clarke Duncan), to survey Lucy, expecting to witness criminal dealings. Little do they know that Lucy has simply been set up on a blind date by her chief henchman and best friend, Scud (Jimmi Simpson) to meet an ex-KGB assassin named Ninotchka Kaprova (Jessica Cauffiel).
