Deux minions travaillent dans le labo de Gru. La pause déjeuner est encore loin mais l'un des minions a une terrible envie de manger sa banane ("Banana" en langage minion). Finalement, il prend sa banane et s'apprête à la dévorer, mais son collègue l'envie et veut la banane aussi. Alors qu'ils se battent, leur supérieur arrive et les sépare, mais quand il voit la banane, il ne peut résister à la tentation de la manger. Les trois minions se battant se retrouvent dans une pièce où des bombes sont surélevées et transportés ailleurs. Deux minions se font attraper et montent jusqu'au plafond, et il en va de même à la banane que voulait garder le supérieur. Ce dernier se rend au poste de commande et approche la banane de lui. Voulant l'avoir, il la fait lâcher, mais les bombes sont lâchées elles aussi. Les trois minions se réfugient dans une autre salle de travail ... remplie de minions. Ainsi, de trois on passe à une cinquantaine de minions voulant manger la banane. En appuyant dessus, un des minions projette la chair de la banane en l'air. La chair retombe dans un trou profond. À ce moment, un autre minion arrive et mange une pomme ("Papple" en langage minion.). Tous les autres se ruent sur lui pour lui voler la pomme.

A mysterious vehicle using a huge magnet steals a highly potent mutagen known as PX-41 from a secret laboratory in the Arctic Circle. The Anti-Villain League (AVL) tries to recruit former supervillain, Gru, to find who stole the mutagen, but Gru refuses, claiming he is a legitimate businessman now as well as the father of Margo, Edith, and Agnes. Dr. Nefario, Gru's friend and assistant, has decided to leave him for new employment, claiming he "missed being evil." Gru reluctantly partners with undercover AVL agent Lucy Wilde, and they search a local mall, the Paradise Shopping Mall, disguised as bakery employees. Mexican restaurant owner, Eduardo Pérez, is suspected by Gru of being "El Macho", a legendary supervillain who supposedly died after skysurfing a shark into the center of an active volcano. Gru and Lucy break into Eduardo's restaurant at night, but find nothing to prove that he is the culprit. Meanwhile, Agnes expresses her wish to have a mother someday and believes that Gru will fall in love with Lucy, but despite her excitement, Gru tells her that his relationship with Lucy is strictly professional.

Minions are small, yellow creatures who have existed since the beginning of time, evolving from single-celled organisms into beings who exist only to serve history's most villainous masters. They find great masters—including a T-Rex, a caveman, an Egyptian pharaoh, Dracula, and Napoleon,—but after accidentally killing nearly all of them with their incompetence, they decide to isolate themselves from the world and start a new life in Antarctica. The film begins at 62 West Wallaby Street on Gromit's birthday at breakfast. After being tipped out of bed and dressed using several mechanical contraptions, Wallace is greeted with a large pile of bills. Wallace remembers Gromit's birthday and presents Gromit with a somewhat unwelcome gift of a spiked dog collar, and a second present of a pair of ex-NASA robotic "Techno Trousers", acquired by Wallace to alleviate the burden of taking Gromit for walks. While Gromit is out on a "walk", Wallace realizes they are in financial difficulty and decides to let the spare bedroom out. He is answered by an inscrutable-looking penguin named Feathers McGraw. The penguin comes to stay at the house, pushing Gromit out of his comfortable bedroom and keeping him awake at night with loud music. On the other hand, Wallace takes a liking to him. Feathers also takes an interest in the Techno Trousers after seeing Gromit use their suction feet to walk on the ceiling.

Scooby-Doo and the gang travel to Atlantic City, where Fred enthusiastically takes them to see The Brancusi Circus. They arrive to find that the circus does not open until tomorrow, but decide to look around when they notice that the front door is unlocked. Whilst searching, they meet a man who is pursuing a werewolf that has intruded into the circus. The werewolf attacks the gang, but is eventually scared off by Velma, using a spotlight. The man thanks the gang and introduces himself as Marius Brancusi, the ringleader. Marius inherited the circus from his uncle last year and has been working to modernize it. He informs the gang that the werewolf they had encountered has been shadowing the circus for a few months, has scared off many of his artists, and has stolen jewelry wherever they are. He even suspects that the werewolf might be one of his own artists. Velma notices that this incident alludes to a case during the 18th century, in Ingolstadt, where a werewolf named Hans collected a combination of jewels which enabled him to change into a werewolf at any time. After the gang tells Marius that they solve mysteries, Marius hires them to go undercover as circus performers.

A reporter named Kathy Lane comes to Theodor Geisel's home in order to do a report on the famous Dr. Seuss, where she meets a strange character. When Kathy asks to use him as a source, he reveals himself to be the Cat in the Hat. Curiosity allows her to open a magical book labeled "Open a book, open up your imagination", which pulls her into the world of Dr. Seuss. The Cat in the Hat character shows Kathy a door which leads to a beach. On the beach, they read The Sneetches. Kathy then falls into a kitchen where she meets Mr. Hunch from Hunches in Bunches. They eat lunch and read McElligot's Pool.
