The House of Lords is a grocery shop that sits on the South Bank of the river Thames close to the site of Festival Hall which is noisily under construction. It is owned by the Lord family, a husband and wife with several children. After the Second World War, some British soldiers are guarding a theatre in Germany containing various refugees and prisoners trying to work out what to do with them. In juvenile court, 15-year-old Elizabeth (Pauline Hahn) recounts her story (in flashback) before the judge. In Edwardian London, Professor Henry Higgins (Rex Harrison), a scholar of phonetics, believes that the accent and tone of one's voice determines a person's prospects in society. In Covent Garden one evening he boasts to a new acquaintance, Colonel Hugh Pickering (Wilfrid Hyde-White), himself an expert in phonetics, that he could teach any person to speak in a way that he could pass them off as a duke or duchess at an embassy ball. Higgins selects as an example a young flower seller, Eliza Doolittle (Audrey Hepburn), who has a strong Cockney accent. Eliza's ambition is to work in a flower shop, but her thick accent makes her unsuitable. Having come from India to meet Higgins, Pickering is invited to stay with the professor. The following morning, Eliza shows up at Higgins' home, seeking lessons. Pickering is intrigued and offers to cover all expenses if the experiment should be successful.

In 1945, as a truckload of war survivors stops in front of an Amsterdam factory at the end of World War II, Otto Frank (Joseph Schildkraut) gets out and walks inside. After climbing the stairs to a deserted garret, Otto finds a girl's discarded glove and sobs, then is joined and comforted by Miep Gies (Dodie Heath) and Mr. Kraler (Douglas Spencer), factory workers who shielded him from the Nazis. After stating that he is now all alone, Otto begins to search for the diary written by his youngest daughter, Anne. Miep promptly retrieves the journal for Otto, and he receives solace reading the words written by his young daughter three years earlier.

In contemporary London, Christopher Isherwood attends a literary party for the launch of a memoir, the author of which he is surprised to learn is Sally Bowles. This knowledge sparks a reverie and the film flashes back to Berlin, New Year's Eve 1931. Broke and frustrated with his writing, Christopher plans to spend the night in but his would be gigolo friend Fritz insists they go to a night club to see Fritz's new inamorata, Sally Bowles, perform. Fritz hopes to live off Sally's earnings as a film star but his ardor quickly cools at the sight of her fiancé Pierre, with whom she plans to leave for Paris that night. Instead, Pierre absconds with her money. Chris, taking pity on her, invites her to stay at his boarding house. They arrange for Chris to move to a smaller room and for Sally to take his old room. Over the course of a long and unproductive winter in which Chris cannot write and Sally finds no work, Chris attempts to initiate a sexual relationship with Sally. She rejects him, saying it would spoil their friendship.
