Connie (Nia Vardalos) and Carla (Toni Collette) are two performers whose lifelong friendship and co-obsession with musical theater have brought nothing but career dead ends. Despite this they continue their optimism, hosting a variety act at an airport lounge. After accidentally witnessing a mafia hit in Chicago, they go on the run, landing in Los Angeles. Initially working at a beauty salon, they wind up posing as drag queens and auditioning to host a drag revue at a gay club.
The plot begins in a convenience store where two FBI agents and brothers, Kevin Copeland and Marcus Copeland (Shawn and Marlon Wayans), try to capture members of an organization that sells drugs inside ice cream boxes, posing as dominican clerks. Unfortunately, the first arrival turns out to be a genuine ice cream delivery, and the actual drug dealers manage to get away. The situation is worsened by the fact that Kevin and Marcus have decided to resolve this bust by themselves.

The film opens with Angela Arden kneeling in front of her twin sister Barbara's grave. Angela is a lounge singer who is attempting to resuscitate her floundering career, which became obsolete around the same time Barbara committed suicide. She's unhappily married to her film director husband Sol Sussman, with whom they have two children–Lance, who is gay and emotionally disturbed, and Edith, a "daddy's girl" who is openly contemptuous of her mother. Also living in the house is the snoopy maid Bootsie, who is infatuated with Sol. Bored and unhappy, Angela begins cheating on her husband with Tony Parker, a tennis-playing "lothario" and failed actor who is reputed to be well endowed.
The award-winning film, loosely based on Barrie's work, presents the characters with a twist. It features Peter Pan as an older, androgynous teen, whereas the fairy Tinker Bell is a drugged and burnt out girl, the Lost Boys are hot guys and pot heads, the Darling children are adopted, Wendy Darling is black, Captain Hook is a gay leather man and Tiger Lily is a transvestite. The events take place in Neverland which is an amusement park in the film, rather than the imaginary island.
One Christmas Eve in present-day Tokyo, three homeless people — Gin (ギン), a middle-aged alcoholic, Hana (ハナ), a trans woman and former drag queen, and Miyuki (ミユキ), a runaway girl — discover an abandoned newborn while looking through garbage. Deposited with the unnamed baby is a note asking the finder to take good care of her and a bag containing clues to the parent's identity. The trio sets out to find the baby's parents. The baby is named Kiyoko (清子), literally meaning "pure child" as she is found on Christmas Eve.
The film centers on the experiences of several teenagers at Camp Ovation, a summer theatre camp. The campers are a mishmash of youngsters from different backgrounds and dealing with different issues, but all feel most comfortable while performing. School is out, and the campers reunite as they prepare to board the buses to Camp Ovation. Shy, self-conscious Ellen Lucas happily greets her friends, recovering from an embarrassing situation where she desperately tried to land a prom date, and ended up asking her brother. Nerdy Fritzi Wagner attempts to befriend icy diva Jill Simmons, but ends up toting her luggage instead. Returning camper Jenna Malloran laments that her parents forced her to have her jaw wired shut in order to lose weight. Upon arrival to Camp Ovation, the kids settle in. Ellen converses with her friend Michael Flores, a gay teenager who was violently beaten by his classmates after showing up to his junior prom in drag. The campers are struck dumb with the arrival of Vlad Baumann, a handsome new camper who is, as a staff member marvels, "an honest-to-god straight boy.
Neelakantham Naidu (Prakash Raj) is a big factionist. He has a loyal confidant Narasimha (Suman), who is willing to give his life for the sake of his master. After many years, Neelakantham's wife (Pragathi) gives a birth to a baby girl. But all the earlier babies born to Neelakantham died due to some unknown reason. The priests advise Neelakantham to name his daughter as Gangotri. They also warn Neelakantham that the child has 'jalagandam', hence they ask Neelakantham to take Gangotri to the holy place of Gangotri in the Himalayas and perform 'abhyangana snanam' (bathe with turmeric, milk, etc.) for the next 15 years. After 15 years, Gangotri would be free of any danger. When nobody could entertain the child Gangotri, Simhadri, the son of Narasimha entertains her with a song. Simhadri becomes a servant and friend to Gangotri. They grow up together. Simhadri (Allu Arjun) and Gangotri (Aditi Agarwal) are good friends. In the mean time, Simhadri's mother (Seetha) sacrifices her life to save Neelakantham. When Gangotri attains puberty, Gangotri's paternal aunt (Telangana Shakuntala) restricts Simhadri from meeting Gangotri as it would be wrong. Gangotri and Simhadri realise that they are in love. When Simhadri comes to Gangotri's place clandestinely to meet her, they are caught by Neelakantham. Neelakantham beats Simhadri. Meanwhile, Narasimha comes to the spot and interferes. This causes a rift between Neelakantham and Narasimha. Neelakantam is misinformed about Narasimha's intentions; hence he has bombs planted in the house of Narasimha. Simhadri is away at that time. When Simhadri comes to know that Neelakantam is responsible for the bombing, he comes to Neelakantam's house and challenges that he will marry Gangotri in 365 days. Then Simhadri goes to the holy place Gangotri and waits for his ladylove to come there for the final sacred bath. The rest of the film is about how Simhadri and Gangotri unite.
Zhan Zhao (Andy Lau) is a court officer who learns of a plot to assassinate Judge Bao (Anthony Wong). While on holiday he meets a young man named Bai (Cecilia Cheung) who turns out to be a woman. Zhan Zhao tries to recruit Bai to help him stop the assassination of Judge Bao.
Jamal Jeffries (Miguel A. Núñez, Jr.) is a UBA (fictional version of the NBA) basketball star whose undisciplined on-and-off-court antics have earned him a bad reputation in the basketball community. Jamal is dropped from his team, the Charlotte Beat and suspended indefinitely after he strips naked in protest of being taken out of a game. His agent, Lorne Daniels (Kevin Pollak), is unsuccessful at finding him a new team, and cuts Jamal as a client. As a result, Jamal's life goes downhill: his endorsements drop him, he ends up bankrupt due to lavish spending, his belongings are repossessed, his mansion is foreclosed on, and his girlfriend Tina (Lil' Kim), who only put up with his antics for his money and fame, walks out on him.
The story starts out with the regular lives of three friends—Dave (Barry Watson), Adam (Michael Rosenbaum), and Doofer (Harland Williams)—who are head of the Social Committee in a frat house called KOK (Kappa Omicron Kappa). The KOK are known for throwing heavy parties and socially discriminating against the members of the sorority DOG (Delta Omicron Gamma), who regularly protest the actions of the KOK.
During World War II the British army is attempting to retrieve an Enigma machine from Germany. Having failed in previous attempts they decide to send four men undercover to the factory that makes the devices, deep in Berlin. Unfortunately the factory is populated entirely by women, and they only have men to send. American O'Rourke (LeBlanc), British transvestite Tony Parker (Izzard), genius Johnno (David Birkin) and the reluctant Archie (James Cosmo) are sent to infiltrate the factory dressed as women.
