A mysterious vehicle using a huge magnet steals a highly potent mutagen known as PX-41 from a secret laboratory in the Arctic Circle. The Anti-Villain League (AVL) tries to recruit former supervillain, Gru, to find who stole the mutagen, but Gru refuses, claiming he is a legitimate businessman now as well as the father of Margo, Edith, and Agnes. Dr. Nefario, Gru's friend and assistant, has decided to leave him for new employment, claiming he "missed being evil." Gru reluctantly partners with undercover AVL agent Lucy Wilde, and they search a local mall, the Paradise Shopping Mall, disguised as bakery employees. Mexican restaurant owner, Eduardo Pérez, is suspected by Gru of being "El Macho", a legendary supervillain who supposedly died after skysurfing a shark into the center of an active volcano. Gru and Lucy break into Eduardo's restaurant at night, but find nothing to prove that he is the culprit. Meanwhile, Agnes expresses her wish to have a mother someday and believes that Gru will fall in love with Lucy, but despite her excitement, Gru tells her that his relationship with Lucy is strictly professional.

In the 2030s, the crew of Ares III, a manned mission to Mars, has established a temporary artificial habitat, called the Hab, on Mars, where they are to stay for two months and depart back to Earth in their orbital probe. Eighteen days into the Mars mission, however, a massive Martian storm hits the base and the crew is forced to abandon the planet and launch themselves because of the danger that their escape vehicle will collapse and leave them stranded. While evacuating, astronaut Mark Watney is lost and presumed dead as his biomonitor is damaged. Gru, a supervillain, has his pride injured when an unknown supervillain steals the Great Pyramid of Giza, an action that is described by his colleague Dr. Nefario as "making all other villains look lame." Gru decides to do better, with the assistance of Dr. Nefario, by shrinking and stealing the Moon, an idea based on his childhood dream of being an astronaut, which was always disparaged by his mother Marlena. Walter Mitty is a negative assets manager at Life magazine who daydreams of adventures and has a crush on a coworker named Cheryl. Mitty works with photojournalist Sean O'Connell, whose images are highly regarded. O'Connell has sent Mitty his latest negatives and a wallet as a gift in appreciation of Mitty's work. O'Connell believes negative #25 captures the "quintessence" of Life and that it should be used for the cover of the magazine's final print issue as it converts to online status. The negative is missing, however, and Walter is forced to stall for time with corporate transition manager Ted Hendricks, who is handling the downsizing. While viewing the other negatives outside Life's offices, Cheryl approaches Mitty and suggests that he think of the negatives as clues to Sean's location. They look at three of them, including one of a person's thumb with a unique ring on it, and another of a curved piece of wood. A third picture of a boat leads Mitty to determine that O'Connell is in Greenland. Mitty promptly flies there to find him.

Graeme Willy (Simon Pegg) and Clive Gollings (Nick Frost) are British comic book enthusiasts and best friends who travel to the United States to attend the annual San Diego Comic-Con International, and to take a road trip through the American Southwest to visit sites significant to UFO lore.
