Micky Ward (Mark Wahlberg) is an American welterweight boxer from Lowell, Massachusetts. Managed by his mother, Alice Ward (Melissa Leo), and trained by his older half-brother, Dicky Eklund (Christian Bale), Micky has not had a particularly successful career: He's become a "stepping stone" for other boxers to defeat on their way up. Complicating matters, Dicky, a former boxer whose peak of success was going the distance with Sugar Ray Leonard in 1978 (before Leonard became a world champion), has fallen apart since then, becoming addicted to crack cocaine. He is now being filmed for an HBO documentary he believes to be about his "comeback".

Puppies from Washington become trapped in a truck heading to Ferntiuktuk, Alaska. Upon arrival, the Buddies meet Shasta, a young husky whose 11-year-old owner is determined to win the Alaskan sled dog race. The puppies decide to help Shasta pursue his dreams. Josh is off to his first year of college and Buddy has stayed behind with Josh's little sister, Andrea, and the rest of the family. Andrea, attempting to fit in with her junior high classmates, Andrea decides to join the baseball team and along the way discovers that Buddy also has the uncanny ability to play baseball. Just as the season is settling in, a terrible discovery is made - Buddy's puppies have mysteriously started disappearing with the help of the kidnappers' little helper, Rocky Raccoon. It turns out the kidnappers were researchers who were kidnapping the puppies because they thought they had a special gene that would enable them to play sports. Teenager Josh Framm's mother, Jackie, has just married her veterinarian boyfriend, Patrick Sullivan. Several years after becoming Buddy's legal owner, Josh Framm (Kevin Zegers), now a teenager, becomes angry when his mother, Jackie, begins dating Patrick Sullivan, the town's new veterinarian after a couple of failed dates. It all starts when Sullivan innocently tosses Josh's basketball-savvy dog, Buddy, a football one day, and he discovers that Buddy also has an uncanny ability to play the sport of football. Soon enough, Buddy begins playing on Josh's Junior High football team. Meanwhile, two Russian siblings by the names of Natalya and Popov kidnap Buddy in hopes of having him perform as the special attraction in the Russian circus while Josh runs away when Patrick proposes to his mother. His coach finds him and convinces him that just because Patrick is in his life now, he doesn't have to stop loving his father and he returns home, but Patrick is gone and Buddy is missing. The Timberwolves are forced to play the final game without Buddy and are unable to win. Buddy and the other animals manage to escape and Natalya and Popov are placed into the custody of the Russian embassy after their van falls into a lake following a chase sequence. Meanwhile, Patrick finds Buddy and takes him to the game. With the help of Buddy, the team catches up, but Buddy is tackled and taken out of the game. The Timberwolves are forced to finish the game without him and thanks to Josh and Tommy, they win. Later, Josh stops Patrick from leaving and convinces him to stay. The family later goes to a Seattle Seahawks football game and Buddy sneaks onto the field.

The film opens with an unsuccessful professional clown, Norman F. Snively, and his dog by the name of "Old Blue," doing a show at a birthday party. Due to the dog causing trouble at the birthday party and both being tossed out of the house, Snively angrily takes him in a kennel to a dog pound, until the kennel falls off his truck. The dog is homeless until he meets a 12-year-old boy, Josh Framm (whom the plot mainly revolves around). After the death of his father, who died in a plane crash during a test flight (indicated in the headline of a framed newspaper article briefly read by Josh), Josh moves with his family from Virginia to Fernfield, Washington. Due to heartbreak over his father's passing, he is too shy to try out for his middle school's basketball team and to make any friends. He instead becomes the basketball team's manager, an absurd offer by coach Joe Barker which he accepts. He practices basketball by himself in a makeshift court that he sets up in a disused allotment, where he first meets the dog, naming him Buddy. Josh soon learns that Buddy has the uncanny ability to play basketball.
