En 1873, au Texas, Prescott Roe, un vieil homme voit son fils, Talbot, plonger dans la folie avec le décès de sa femme, Awbonnie, une métisse indienne de la tribu des kiowas. Celle-ci est morte en voulant mettre au monde un enfant et Talbot ne s'en remet pas. Prescott désespère de ne pas pouvoir lui venir en aide. Il décide alors d'aller rencontrer le père de la jeune femme défunte, Eamon McCree, un shaman charlatan qui a un fort penchant pour l'alcool et qui avait vendu sa fille en échange de trois chevaux. The story focuses on the Magrath sisters—Lenny, Meg, and Babe—who reunite at the family home in Hazlehurst, Mississippi after Babe shoots her abusive husband. The three were raised by Old Granddaddy after their mother hanged herself and the family cat and have been eccentric ever since. Lenny is a wallflower who bemoans her shriveled ovary. Egocentric Meg is a singer whose Hollywood career ended abruptly when she suffered a nervous breakdown. Unruly and impulsive Babe shocks her sisters with stories about her affair with a teenaged black boy. Michael Dorsey (Dustin Hoffman) is a respected but perfectionist actor. Nobody in New York wants to hire him anymore because he is difficult to work with. According to his long-suffering agent George Fields (Sydney Pollack), Michael's attention to detail and difficult reputation led a commercial he worked on to run significantly over-schedule, because the idea of a tomato sitting down was "illogical" to him. After many months without a job, Michael hears of an opening on the soap opera Southwest General from his friend and acting student Sandy Lester (Teri Garr), who tries out for the role of a hospital administrator Emily Kimberly but does not get it. In desperation, and as a result of his agent telling him that "no one will hire you", he dresses as a woman, auditions as "Dorothy Michaels" and wins the part. Michael takes the job as a way to raise $8,000 to produce a play, written by his roommate Jeff Slater (Bill Murray) and to star Sandy, titled Return to Love Canal. Michael plays his character as a feisty, feminist administrator, which surprises the other actors and crew who expected Emily to be (as written) another swooning female in the plot. His character quickly becomes a television sensation.

Don Anderson is the Mickey's hamburger chain marketing director who helped develop the "Big One", its most popular menu item. When he learns that independent research has discovered a considerable presence of fecal matter in the meat, he travels to the fictitious town of Cody, Colorado to determine if the local Uni-Globe meatpacking processing plant, Mickey's main meat supplier, is guilty of sloppy production. Ritter plays Ed Chandler, a father who is fired from his job for taking time off to tend to his 16 year daughter Missy (Chlumsky), who is battling cancer. Rather than accepting his dismissal, Ed decides to fight back. With the help of a United States senator, he lobbies the United States Congress and is the stimulus to passage of the Family and Medical Leave Act. Because Missy's cancer is terminal, she is visited by the Make-A-Wish Foundation and she wishes to go to the White House and visit the Oval Office and hopefully see President Bill Clinton himself. Although Make-A-Wish is not so sure if they can grant her wish, in the end she does in fact get to visit the Oval Office and meet President Clinton himself in a surprise cameo appearance.

At the deathbed of her wealthy cousin Adeline (Chaplin), poor seamstress Bette Fischer (Lange) promises to take care of her cousin's family. Bette finds a source of comfort in Count Wenceslas Steinbach (Aden Young), a young but promising artist who has fallen on hard times. Besotted by the young man, Bette does everything possible to provide for him such as buying him food and motivating him to continue with his work. One day, Bette tells Hortense of her "sweetheart" Wenceslas.
