L'histoire se déroule cinq ans après les événements de Sharknado 3. La tempête porteuse de requins tueurs est de retour et la cible des squales est à présent Las Vegas. Après avoir ravagé la ville, le Sharknado se déplace et frappe plusieurs états du centre des États-Unis, sans façade maritime, qui n'ont pas l'habitude des cyclones se formant au-dessus de la mer, mais plutôt des tornades continentales. En se déplaçant et frappant diverses installations (barrage Hoover, champ de pétrole, transformateur électrique, centrale nucléaire), la tornade change de nature en se chargeant de matières de plus en plus dangereuses : poussières, pierres, pétrole, feu, électricité à haute tension, et même radioactivité. Toute la famille Shepard, enfin réunie, va devoir unir ses forces pour en venir à bout.

The story takes place at East Great Falls High, ten years after the events of American Pie. The new protagonists of this movie are three brand new hapless virgins: Rob (Hall), Nathan (Horton), and Lube (Hardesty). One night Rob accidentally sets fire to the school library, and finds "the Bible" (or Book of Love), the creation of Mr. Levinstein (Eugene Levy). Unfortunately for them, the book is ruined. The book had incomplete advice and sends them on a helpless journey to lose their virginity. To celebrate his upcoming marriage to Tracy Garner (Sasha Barrese), Doug Billings (Justin Bartha) travels to Las Vegas with his best friends Phil Wenneck (Bradley Cooper) and Stu Price (Ed Helms), as well as Tracy's brother and Doug's future brother-in-law Alan (Zach Galifianakis), in Doug's future father-in-law's vintage Mercedes-Benz. They stay at Caesars Palace, where they relax in the room before celebrating with a few drinks on the hotel rooftop. The next morning, Phil, Stu, and Alan awaken to find they have no memory of the previous night, and Doug is nowhere to be found. Stu is missing a tooth, their hotel suite is in disarray, a tiger is in their bathroom, a chicken in their living room, and a baby is in the closet, whom they name "Carlos". They find Doug's mattress impaled on a statue outside of their hotel and when they ask for their Mercedes, the valet delivers an LVPD police cruiser.

The movie begins with Eugene (Zach Cregger) and his best friend Tucker (Trevor Moore) as kids. They sneak into the bedroom closet of Eugene's brother, as he recently acquired a valuable Michael Jordan rookie card. Upon opening the box the card was in, Tucker notices that Eugene's brother has an issue of Playboy. Tucker opens the magazine, instantly becoming obsessed with it. Seth (Jonah Hill) and Evan (Michael Cera) are two high school seniors who lament their virginity and poor social standing. Best friends since childhood, the two are about to go off to different colleges, as Seth didn't get accepted into Dartmouth like Evan. After Seth is paired with Jules (Emma Stone) during Home-Ec class, she invites him to a party at her house later that night. Later, Fogell (Christopher Charles Mintz-Plasse) comes up to the two and reveals his plans to obtain a fake ID during lunch. Seth uses this to his advantage and promises to bring alcohol to Jules' party. Fogell's fake ID is met with derision by Seth and Evan, as it states that Fogell's name is simply "McLovin". After contemplating their options, Seth decides they have no choice but to have Fogell buy the alcohol with his fake ID. Fogell goes in and successfully buys the alcohol, but is interrupted when a robber enters the store, punches him in the face, and takes money from the cash register.
