The film begins with GloboTech Industries acquiring the Heartland Toy Company. CEO Gil Mars tells remaining toy designers Larry Benson and Irwin Wayfair to develop actual live-action toys capable of "playing back". Mars selects Larry's action figures, the Commando Elite, for the project and Irwin's educational toys, the Gorgonites, for their enemies. After Mars sees a commercial showing the toys doing activities they are incapable of doing, he accuses them of false advertising, saying Globotech delivers on what it promises. Mars then orders Benson to have the toys on the shelves in three months acting just as they do in the commercial or get fired. Faced with such a tight deadline, Benson forgoes safety testing, then uses Irwin's password and chooses GloboTech's overly powerful, X1000 intelligent AI munitions microprocessor integrated circuit to control the toys.

Megamind is a super intelligent alien who was evacuated from his homeworld as a baby, as was Metro Man; the two fall into paths of super villainy and superheroism respectively and grow up as rivals fighting for control of Metro City. Megamind is consistently defeated by Metroman and is in prison. After using a holographic watch to escape with the aid of Minion, a talking fish with the robotic body of a gorilla, he kidnaps Metro Man's supposed love interest, reporter Roxanne Ritchi and holds her hostage to lure him into a trap. Finding that copper is Metro Man's one weakness, Megamind's plan to obliterate him with a death ray powered by the sun succeeds, and Megamind finally takes over the city. His joy is short lived though, as without a hero to fight, he finds his life has become meaningless. He goes to the Metroman Museum, which was dedicated to him on the day of his death and nearly runs into Roxanne. He uses his holographic watch to disguise himself as the museum's curator Bernard, and she innocently gives him the idea of creating a new superhero to take Metro Man's place. After creating a formula from Metroman's DNA, Roxanne intervenes in his plans and he accidentally injects the serum into Hal Stewart, Roxanne's dimwitted cameraman, who has an unrequited crush on her. Under the guise of his "Space Dad", Megamind tries to mold Hal into a superhero named Titan (misheard by Hal as "Tighten"), as it was the only name he could trademark. Unfortunately, Hal's ambitions are crushed when he sees Roxanne and Megamind as Bernard on a date. However, Megamind's disguise falters during dinner and she rejects him, causing him to lose track of his invisible car which contains the gun capable of removing Hal's powers.

In 1989, scientist Hank Pym resigns from S.H.I.E.L.D. after discovering their attempt to replicate his Ant-Man shrinking technology. Believing the technology is dangerous, Pym vows to hide it as long as he lives. In the present day, Pym's estranged daughter, Hope van Dyne, and former protégé, Darren Cross, have forced him out of his own company. Cross is close to perfecting a shrinking suit of his own, the Yellowjacket, which horrifies Pym.

Benjamin Jahrvi (Fisher Stevens), now unemployed, is peddling sophisticated toy robots that he makes himself by hand on the street corners of New York City. One robot wanders away from his stand and by chance makes its way into the office of Sandy Banatoni (Cynthia Gibb), a scout for a major toy company. Sandy tracks Ben down and orders 1,000 of his toys. Overhearing this offer is con artist Fred Ritter (Michael McKean), who smooth-talks his way into becoming Ben's business partner in the deal and later acquires the funding Ben needs to get his assembly line started from a loan shark.

Hiro Hamada is a 14-year-old robotics genius in the futuristic fictional city of San Fransokyo. Raised by his aunt Cass and older brother Tadashi after the death of his parents, he spends his time participating in illegal robot fights. To redirect Hiro, Tadashi takes him to the robotics center at his university, where Hiro meets Tadashi's friends: GoGo, Wasabi, Honey Lemon, and Fred, as well as Baymax, the inflatable healthcare companion robot Tadashi created. To enroll in the school, Hiro signs up for the school's science fair and presents microbots: swarms of tiny robots that can link together in any arrangement imaginable. Professor Callaghan, the head of the university's robotics program, is impressed, and grants Hiro entrance to the university. Alastair Krei, renowned entrepreneur and president of Krei Tech, offers to buy the microbots, but Hiro follows Callaghan's warnings about Krei's questionable business practices and declines to sell them. When Tadashi and Hiro later arrive at the university to find it engulfed in flames, Tadashi rushes in to rescue Callaghan, but the building explodes moments later, killing him.

After a decade of research, the scientist Dr. Vaseegaran creates a sophisticated android robot with the help of his assistants, Siva and Ravi, in order to commission it into the Indian Army. He introduces the robot, named Chitti, at a robotics conference in Chennai. Chitti helps Sana, Vaseegaran's medical student girlfriend, cheat in her examination, then saves her from being assaulted by a group of thugs. John Brown is a humble, but clumsy security guard working at the Bradford robotics laboratory in Riverton, Ohio, run by Artemus Bradford and his daughter Brenda, whom Brown has a crush on. Brown wishes to become a police officer, with support from his niece Penny. Brenda and Artemus create a lifelike robotic foot as part of the Gadget Program, to create cybernetically augmented police officers. Tycoon Sanford Scolex attacks the lab, steals the foot, and has Artemus murdered before escaping in his limo. Brown goes chasing him down in his car, which leads to its being destroyed by Scolex's explosive cigar leaving brown for dead, but a bowling ball coming from the fiery blast of Brown’s destroyed car smashingly crushes Scolex's hand, forcing him to replace it with a mechanical claw.
