Military Intelligence officer and former policeman Lt. Col. Daniel Kiley (Fonda) and his pilot, Joe, are flying a reconnaissance mission over the Ardennes forest, spotting a German staff car. Under the ground in a subterranean lair, German Col. Martin Hessler (Shaw), a fictional character loosely based on SS-Standartenführer Jochen Peiper, is briefed by his superior, Gen. Kohler (Werner Peters). Kohler points out a clock with a 50-hour countdown, which is the time allotted for the operation, beyond which Germany has no resources for full-scale attack. At the same time German soldiers disguised as American troops, led by Lt. Schumacher (Ty Hardin), are tasked with seizing vital bridges and sowing confusion behind the Allied lines.

The Great Leslie (Tony Curtis) and Professor Fate (Jack Lemmon) are competing daredevils at the turn of the 20th century. Leslie is the classic hero – always dressed in white, handsome, ever-courteous, enormously talented and successful. Leslie's nemesis, Fate, is the traditional melodramatic villain – usually dressed in black, sporting a black moustache and top hat, glowering at most everyone, maniacal evil laugh, grandiose plans to thwart the hero, and dogged by failure. Leslie proposes an automobile race from New York to Paris, to prove the ability of a new car named after him. Fate builds his own race vehicle, the Hannibal Twin-8, complete with hidden devices of sabotage. Others enter cars in the race, including New York City's most prominent newspaper. Driving the newspaper's car is beautiful photojournalist Maggie DuBois (Natalie Wood), a vocal suffragette.

The West Berlin office of the Circus, under administrator Alec Leamas (Richard Burton), has not been doing well. He is recalled to London shortly after the death of one of his operatives. Leamas is seemingly demoted to the banking section of the agency. In reality, a carefully staged transformation of Leamas has been arranged by Control (Cyril Cusack), the agency's chief. Now depressed and disgruntled, alcoholic and low on funds, he is quickly spotted by the East German Intelligence Service as a potential defector.

Frank Towns (James Stewart) is the pilot of a twin-engine Fairchild C-82 Packet cargo plane flying from Jaghbub to Benghazi in Libya. Lew Moran (Richard Attenborough) is the navigator while the passengers are Capt. Harris (Peter Finch) and Sgt. Watson (Ronald Fraser) of the British Army; Dr. Renaud (Christian Marquand), a physician; Heinrich Dorfmann (Hardy Krüger), a German aeronautical engineer; Mr. Standish (Dan Duryea), an oil company accountant; and several oil workers that include Trucker Cobb (Ernest Borgnine), a mentally-disturbed foreman; Ratbags Crow (Ian Bannen), a mean-spirited, sardonic Scot; Carlos (Alex Montoya) and his pet monkey; and Gabriel (Gabriele Tinti). Poitier portrays Alan, a Seattle college student who is volunteering at Seattle's then-new Crisis Clinic, a crisis call center.
