During the French and Indian War, Alice (Barnes) and Cora Munro, the two daughters of the British commander of a fort, set out from Albany to join him. They are escorted by Major Duncan Heyward (Wilcoxon), who has loved Alice for a long time, and by the Huron Indian Magua. Magua is secretly an enemy of the British. Magua betrays them, but they are rescued by a colonial scout named Hawkeye (Scott) and his friends, the last two members of the Mohican tribe, Chingachgook and his son Uncas. On their way to the fort, Uncas falls in love with Cora, while Hawkeye and Alice are attracted to each other.

A few short hours after the assassination of President Abraham Lincoln (Frank McGlynn Sr.), Dr. Samuel Mudd (Baxter) gives treatment to a man with a broken leg who shows up at his door. Mudd does not know that the president has been assassinated and the man who he is treating is John Wilkes Booth (Francis McDonald). Mudd is arrested for being an accessory in the assassination and is sent to prison on the Dry Tortugas, described as in the West Indies and referred to in the film as "America's own Devil's Island".

Pamela defies her autocratic father (Donald Crisp), and has a baby out of wedlock with her lover, Gerald Waring (Van Heflin, in his screen debut). Pamela raises her illegitimate daughter as her niece after her pregnant sister (Elizabeth Allan) falls and dies over the death of her young husband and becomes a crusading journalist for women's rights. Eventually she agrees to marry diplomat Thomas Lane (Herbert Marshall) after being unfairly named as co-respondent in Waring's divorce. Hepburn's performance as the defiant young woman is considered the epitome of her feminist characterizations of the 1930s.

Nord du Japon, vers 1670. Sous le nom d'Akanishi Kakita, un homme au physique disgracieux aimant son chat et le shōgi est en réalité un espion chargé de surveiller les agissements dans le clan Date pour le compte du chef du clan Katakura. Ayant réussi à déjouer une tentative d'assassinat et intercepter des courriers compromettants, il cherche un prétexte pour quitter la résidence du seigneur Date Hyōbu sans éveiller les soupçons. Son acolyte Aozame Masujirō lui conseille d'écrire une lettre d'amour à Sazanami, la plus belle femme de la résidence Date pour devenir la risée de ses compagnons et ainsi obtenir une justification à son départ. Mais comble de malchance, la belle répond favorablement à ses avances l'obligeant à "oublier" une autre lettre pour qu'elle soit découverte et qu'il puisse profiter du stratagème. Soupçonneux, le seigneur Kai Harada se doute de quelque chose et envoie une troupe à sa poursuite, mais celle-ci échoue à le retrouver.

Son ami Kurata vient à sa rencontre, il pense qu'il craint de retourner dans la ville qu'il a fui huit ans auparavant, en abandonnant sa première femme et son fils Sentarō. Kumoemon Tōchūken passe la nuit à boire puis revoit son fils venu avec Kurata. Il décide au soulagement de tous de repartir le lendemain pour Tokyo.
