In preparation for her birthday celebration, the Smurfs read the story of Smurfette, how she was created by Gargamel to destroy them, only for Papa Smurf to rescue her and turn her fully into a Smurf. Meanwhile, she is having nightmares about reverting to her original form and betraying her fellow Smurfs by turning them over to Gargamel. The Smurfs are preparing a surprise party, but as Smurfette tries to find out what her fellow Smurfs are planning, none of them are saying a word. She takes this to mean that everyone forgot her birthday.

As the Smurfs get ready for the Blue Moon Festival, Papa Smurf sees in his cauldron a vision of Clumsy Smurf reaching for a dragon wand and the Smurfs in cages while Gargamel laughs. Not wanting this vision to come true, Papa Smurf disallows Clumsy to pick Smurf Roots. Clumsy disobeys Papa and ends up unintentionally leading Gargamel and Azrael to the village. The Smurfs all flee for their lives while Clumsy unknowingly runs towards the Forbidden Falls, with Papa Smurf, Smurfette, Grouchy, Brainy and Gutsy running after him. They find him at the edge of a cliff, and while trying to help him up, they are sucked into a gigantic vortex that spirits them to present day New York City. To make matters worse, Gargamel and Azrael follow and the Smurfs end up in the apartment of Patrick and Grace Winslow, a married and expectant couple and their Basset Hound Elway. After clarifying things, the Winslows befriend them and allow them to stay in their apartment. The next day, needing to find a "star gazer" (a telescope), the Smurfs follow Patrick to his work place at Anjelou Cosmetics, misunderstanding the previous explanation of his job as fortune-telling. He calls Grace to pick them up.

Mystery Inc. (Fred, Daphne, Velma, Shaggy and Scooby-Doo) attend the opening of an exhibition at the Coolsonian Criminology Museum commemorating their past solved cases with monster costumes on display. However, the celebrations are interrupted by the Evil Masked Figure who steals two costumes using the reanimated Pterodactyl Ghost. The gang are ridiculed by journalist Heather Jasper Howe who starts a smear campaign against them. Concluding an old enemy is the mastermind, the gang revisit old cases, dismissing the former Pterodactyl Ghost, Jonathan Jacobo, due to his death during a failed prison escape, they guess Jeremiah Wickles, the Black Knight Ghost’s portrayer, is the culprit.

In a warehouse, the Mystery, Inc. gang illustrates a plan to catch the Luna Ghost who has kidnapped Daphne Blake (Sarah Michelle Gellar), flying around with her bound and gagged which goes astray but ends with Shaggy Rogers (Matthew Lillard) and Scooby-Doo (voiced by Neil Fanning) causing the Ghost to be caught. The gang unmasks the Luna Ghost, who is revealed the janitor of the Wow-O-Toy Factory, Old Man Smithers, who wanted revenge after Pamela Anderson refused to go out with him (it is implied that he kidnapped Daphne, mistaking her for Pamela). After solving the mystery, constant arguments among the members of Mystery Incorporated about Fred Jones (Freddie Prinze, Jr.) taking credit for Velma Dinkley's (Linda Cardellini) ideas and Daphne always getting kidnapped cause the gang to go their separate ways, much to the dismay of Shaggy and Scooby.

The film begins in an illegal underground dog-fighting arena in Korea, where an FBI agent named John Maxwell (Paul Giamatti) has been identified. John is ordered to be killed by a Korean mob boss, but is eventually rescued by his undercover partner and master of disguise, Malcolm Turner (Martin Lawrence). Suddenly, a group of FBI agents storm around the arena. Meanwhile, a criminal named Lester Vesco (Terrence Howard), who was originally serving a life sentence in prison for murder and armed robbery, escapes from his cell by killing the doctor and stealing his car. The FBI assigns Malcolm and John to capture Lester by sending them to small-town Cartersville, Georgia to stake out the house of a fat, elderly African American woman named Hattie Mae Pierce (Ella Mitchell; whom her friends call "Big Momma"), the estranged Southern grandmother of Lester's ex-girlfriend, Sherry Pierce (Nia Long), who supposedly aided Lester in his original bank robbery by giving him the key to the vault. After Big Momma unexpectedly leaves town to help her ill friend within a couple of weeks, Malcolm and John sneak into her house to plant security cameras and tap the phones. Sherry later calls Big Momma's house and Malcolm disguises his voice as Big Momma in order to lure Sherry and possibly obtain a confession. The plan works, in which Malcolm and John work together on a Big Momma disguise costume before Sherry's arrival the next day.

Child psychologist Samuel Faulkner (Hugh Grant) has an ideal romance with ballet teacher Rebecca Taylor (Julianne Moore). Rebecca is thinking about marriage and children. Samuel is against the idea of marriage as he is happy with how things are between them. This all changes when Rebecca declares she is pregnant and when questioned by Samuel about her birth control she replies birth control is only 97% time effective. His fears mount due to his encounters with overbearing couple Marty (Tom Arnold) & Gail (Joan Cusack) and their unruly daughters, as well as the confusing advice he gets from his perpetually single artist friend (Jeff Goldblum). They meet their Doctor Kosevich (Robin Williams) who happens to be Russian. Samuel is confused and unsure about what to do. Feeling neglected Rebecca leaves him and moves in with Marty and Gail. When Cole's Department Store's Santa Claus gets drunk before taking part in the Thanksgiving parade, Dorey Walker (Elizabeth Perkins), Cole's director of special events, fires him and must find a replacement immediately. She spots an old man (Richard Attenborough) berating the inebriated Santa, and begs him to take over. He introduces himself as Kris Kringle. Kris does so well during the parade that he is immediately hired to be Cole's main Santa for the holiday period. All the children in New York begin to believe that he is the real Santa, with the exception of Dorey's six-year-old daughter Susan (Mara Wilson). Dorey's boyfriend, Bryan Bedford (Dylan McDermott), does his best to convince Susan to believe. While being babysat one night by Kris, Susan shares with him her Christmas wish, she would like a dad, a house (used every year for the Cole's catalogue photoshoot) and a baby brother. Kris asks if she would begin to believe in Santa if she got all those things. Susan agrees that she would.
