Infuriated at being told to write one final column after being laid off from her newspaper job, Ann Mitchell (Barbara Stanwyck) prints a letter from a fictional unemployed "John Doe" threatening suicide on Christmas Eve in protest of society's ills. When the letter causes a sensation among readers, and the paper's competition suspects a fraud and starts to investigate, editor Henry Connell (James Gleason) is persuaded to rehire Mitchell, who schemes to boost the newspaper's sales by exploiting the fictional John Doe. From a number of derelicts who show up at the paper claiming to have written the original letter, Mitchell and Connell hire John Willoughby (Gary Cooper), a former baseball player and tramp in need of money to repair his injured arm (by Bonesetter Brown), to play the role of John Doe. Mitchell starts to pen a series of articles in Doe's name, elaborating on the original letter's ideas of society's disregard for people in need.

In 1906, Alipang (Tetsu Komai) and his Muslim Moro guerrillas are terrorizing the people of the Philippine island of Mindanao, raiding villages, killing the men, and carrying off the women and children for slaves. Instead of maintaining garrisons indefinitely to protect the Filipinos, the U.S. army tests out a new tactic at Fort Mysang. The army detachment is replaced by a handful of officers – Colonel Hatch (Roy Gordon), Captains Manning (Russell Hicks) and Hartley (Reginald Owen), and Lieutenants McCool (David Niven) and Larsen (Broderick Crawford) - who are to train the native Philippine Constabulary to take over the burden. Amid vignettes involving a philandering husband who fakes amnesia, a child prodigy who yearns to live a normal life, an aging bachelor whose housekeeper has disappeared, and an old lady whose daughter has run away, the primary plot line focuses on brash detective Butch Saunders, who is assigned to find missing Chicago banker Therme Roberts. Butch finds himself attracted to the man's wife Norma, despite the fact they're both married, so when his superior, Captain Webb, tells him she really is Norma Phillips and the man she claims is missing is not her husband but the person she has been accused of murdering, he doesn't believe her. Norma fakes her suicide by drowning and disappears, but can't resist returning when Butch stages her funeral in the hope she'll surface. Not only she but the missing Roberts, as well, turn up at the services. Norma tells Butch she once was Roberts' secretary, and he killed his mentally disturbed twin brother and assumed his identity in order to avoid embezzlement charges. Roberts denies her accusations, but Webb tricks him into admitting his guilt. Norma is cleared and, when Butch learns his wife Belle never divorced her first husband, the two are free to wed.
