James (Peck), at the age of 27 quits his law career in pursuit of his childhood dream of becoming an actor. While studying acting under the tutelage of Shelley (Harden), he becomes engrossed in Carlo Lucchesi's play, Tinto de Verano, which is set in Spain. James’ girlfriend Brit (Chow) leaves him, and he spontaneously flies to Spain, where he encounters the playwright Lucchesi (Wells) at a bookstore in Barcelona. Mexican filmmaker Sebastián (Gael García Bernal) and his executive producer Costa (Luis Tosar) arrive in Cochabamba, Bolivia, accompanied by a cast and crew, prepared to create a film depicting Columbus's first voyage to the New World, the imposition of Columbus’ will upon the natives, and the subsequent indigenous rebellion. Cognizant of his limited budget, producer Costa elects to film in Bolivia, the poorest country in South America. There, impoverished locals are thrilled to earn just two dollars a day as extras in the film, and willingly engage in physical labor for set preparation. Costa saves many thousands of dollars by having underpaid extras perform tasks meant to be completed by experienced engineers.

The film opens on a ferry in Liverpool, as Fergus Molloy (Mark Womack) remembers the final messages sent to him by his lifelong friend Frankie (John Bishop), whose funeral he is to attend. The night before, Molloy unseals his friend's coffin to see his friend's badly injured corpse. At the funeral, Haynes (Jack Fortune) a director of the private military company that Molloy and Frankie worked for, gives a eulogy praising Frankie and describing military contractors as the "unsung heroes of our time". Afterwards, Haynes and Walker (Geoff Bell) explain to Frankie's family the circumstances of his death, though Molloy remains embittered and closely questions the two men. Later at the wake, Molloy attacks Haynes when he sees him distributing his business card to enlisted soldiers there.

Inés, Eva and Carmen, three women of different generations, work as private detectives for an investigation agency run by Valbuena, their male chauvinist boss. Before Night Falls is based on the autobiography of the same name by Cuban poet and novelist Reinaldo Arenas. In the film, Arenas, who was openly gay, is born in Oriente in 1943 and raised by his single mother and her parents, who soon move the entire family to Holguín. After moving to Havana in the sixties to continue his studies, Reinaldo begins to explore his ambitions, as well as his sexuality. After receiving an honorary mention in a writing contest, Arenas is offered the chance to publish his first work. Through his work and friendships with other openly gay men (such as Pepe Malas and Tomas Diego), Arenas manages to find himself.
